Are Weight-Loss Drugs Also Longevity Drugs?

Two of the hottest trends in health — longevity medicine and weight-loss drugs — are colliding.

A theory that GLP-1 medications like Ozempic or Zepbound might help people live longer has been bandied about by biohackers and aging researchers for several years. Online pharmacies already market compounded versions of the drugs to purportedly extend health span. But so far, academic research into the hypothesis has been scant.

One of the first studies putting the idea to the test was published last month. It showed that, among people with H.I.V. and lipohypertrophy (fatty deposits that develop under the skin), taking semaglutide (the drug in Ozempic) for eight months seemed to slow participants’ biological aging, according to blood tests that measure age-related biomarkers. People with H.I.V. experience accelerated aging because of the infection, so they are a good population in which to conduct age-related research, said Michael Corley, an associate professor of medicine at the University of California, San Diego’s Stein Institute for Research on Aging, who led the study.

The trial was preliminary, but it “provided us an opportunity to say, hey, is there any signal here that warrants all the hype?” Dr. Corley said.

That hype stems from several lines of evidence. First, there is ample research demonstrating that GLP-1s improve people’s metabolic health by helping to regulate insulin and blood sugar levels and causing weight loss. Several studies show that they also benefit cardiovascular, liver and kidney health.

This is really long ... sorry

I was 56, morbidly obese, bad blood chemistry, high blood pressure, muscle atrophy ... a walking metabolic disaster

I still had 9 years to go before retirement, and I realized I was not going to make it

I started a new program based on Intermittent Fasting, walking and weight lifting ... It worked; I lost 110#, saved my life and my retirement

During this phase, I started learning of the extraordinary benefits of fasting, and especially, I learned about glucagon

I had never heard of glucagon before

It is produced by the pancreas, and only released when blood glucose approaches zero ... it switches your body from using dietary blood glucose for energy to extracting triglycerides from lipocytes and converting triglycerides to glycogen (glucose)

Insulin causes glucose to be stored in lipocytes

Glucagon causes glucose to be released from lipocytes

These two hormones form the basic controls for our energy storage system

But, there is more to glucagon than just controlling the flow of energy to and from our fat cells: It only starts after we've exhausted our supply of blood sugar, and enter a state of starvation.

We acquire macronutrients from our meals: carbs for blood glucose, fats for essential lipids related to cell membranes and certain energy processes, and proteins, which are used to rebuild/restore the body's tissue cells.

When we exhaust these macronutrients, the body has a system of alternatives, which will kick into gear at starvation:

The first is the return of stored glucose in the fat cells. Glucagon opens fat cells to allow the release of stored energy by converting those stored triglycerides to glycogen/glucose: This is how we lose weight

The lack of protein during starvation is another critical problem, and so the body has evolved a system of cellular degradation and recycling known as autophagy

Our bodies are constantly recycling tissue cells through regular autophagy, known as cellular turnover. Hundreds of millions of cells are repaired every second.

However, during starvation, there is little protein remaining to use as spare parts for cellular turnover, so, the presence of glucagon triggers the stem cells around the body to accelerate autophagy, to find and remove defective cellular components, to break those components down to it's constituent amino acids, in lysosomes, and use those amino acids to replace the missing proteins for continued cellular turnover, so cells can still be repaired during starvation

Many believe that part of aging is an accumulation of damaged cells that go unrepaired ...

Starvation stimulates accelerated autophagy to 500% of basal autophagy when we are in the fed state ... That's 500% more cellular defects repaired ...

So, many of those who fast are purposely causing accelerated autophagy in order to reach the 500% rate of autophagic turnover

More cells repaired = less damage from aging, or so the thinking goes

So, when they created this 'Glucagon-like peptide', I wondered how this version would work in the human body, and would it also cause accelerated autophagy like natural glucagon?

It seems to work to release stored triglycerides/glycogen, which will reduce weight

But does it stimulate accelerated autophagy, and help increase the rate of cellular repair like natural glucagon?

There is evidence, like this report shows, that Glucagon-like peptides DO stimulate higher rates of cellular repair, and so would help your body retain healthier cells that are repaired more often

These drugs are like 'fasting in pill form'

I've used a fasting lifestyle to do this naturally, so I get my glucagon from starvation, and that's free

But I will admit; If you can't fast, using Glucagon-like peptides looks to be an easy street to accelerated autophagy, as well as weight loss

Glucagon is so important to our health: Why were they so quiet about this critical aspect of our health?

It was always 'insulin this' and 'insulin that', and nary a word of it's compliment

I was shocked when I realized my ignorance of this information over the decades

So, glucagon AND glucagon-like peptides will help you lose weight AND help with cellular regeneration and cellular health through enhanced autophagy
